Psoriasis is a common and chronic inflammatory skin disease complicated by genetic-environmental interactions. Although genomic, transcriptomic and proteomic analyses have been performed to investigate the pathogenesis of psoriasis, the role of metabolites in psoriasis, particularly of lipids, remains unclear. Lipids not only comprise the bulk of the cellular membrane bilayers but also regulate a variety of biological processes, such as cell proliferation, apoptosis, immunity, angiogenesis and inflammation. <br> In this study, an untargeted lipidomics approach was used to study the lipid profiles in psoriasis and identify lipid metabolite signatures for psoriasis through ultra-performance liquid chromatography-quadrupole tandem mass spectrometry. Plasma samples from 90 participants (45 healthy and 45 psoriasis patients) were collected and detected. Statistical analysis was applied to find different features between the two subjects. In addition, ELISA was performed to examine differential expression lipids in psoriatic patient plasma. We finally identified several differential expression lipids including LPAs, LysoPCs, PIs, PCs and PAs, among these metabolites, LPAs, LysoPCs and PAs were significantly increased, while PCs and PIs were down-regulated in psoriasis group. <br>

银屑病是一种常见的慢性炎症性皮肤病,其发病与遗传-环境交互作用密切相关。尽管目前已通过基因组学、转录组学和蛋白质组学分析对银屑病的发病机制开展了诸多研究,但代谢物尤其是脂质类代谢物在银屑病中的作用仍不明确。脂质不仅是细胞膜双分子层的主要组成成分,还可调控细胞增殖、凋亡、免疫应答、血管生成及炎症反应等多种生物学过程。 本研究采用非靶向脂质组学方法,借助超高效液相色谱-四极杆串联质谱(ultra-performance liquid chromatography-quadrupole tandem mass spectrometry)技术分析银屑病患者的脂质谱,以期筛选出银屑病相关的脂质代谢特征。研究收集并检测了90名受试者的血浆样本,其中45名为健康对照者,45名为银屑病患者,通过统计学分析筛选两组间的差异脂质特征。此外,本研究还通过酶联免疫吸附测定(Enzyme-Linked Immunosorbent Assay,简称ELISA)验证了银屑病患者血浆中差异表达脂质的表达水平。最终本研究共鉴定出多种差异表达脂质,包括溶血磷脂酸(LPAs)、溶血磷脂酰胆碱(LysoPCs)、磷脂酰肌醇(PIs)、磷脂酰胆碱(PCs)与磷脂酸(PAs);其中,溶血磷脂酸、溶血磷脂酰胆碱与磷脂酸在银屑病组中显著上调,而磷脂酰胆碱与磷脂酰肌醇则呈下调趋势。
