Characterization of differential gene expression pattern in wild-type and C3 deficient mice during retinal vascular development

This study reports the comprehensive gene expression profiling of postnatal day (P) 5 wild-type (WT) and complement component (C) 3 mutant (referred as KO) retinas during development. Further functional enrichment analysis revealed up-regulation of biological processes linked to extracellular matrix organization. Retinal mRNA profiles of P5 WT and C3-KO mice were generated by deep sequencing, n=4/genotype, using Illumina Nextseq 2000, and the sequencing target was 25 million reads per samples.

本研究报道了发育过程中出生后第5天(postnatal day 5, P5)野生型(wild-type, WT)与补体成分3(complement component, C3)突变体(下称敲除型KO)小鼠视网膜的全基因表达谱分析结果。进一步的功能富集分析显示,与细胞外基质组织相关的生物学过程出现上调表达。本研究采用Illumina Nextseq 2000测序平台,对P5 WT及C3-KO小鼠的视网膜mRNA表达谱进行深度测序(deep sequencing),每个基因型设置4个生物学重复,单样本测序目标读段数为2500万条。
