The high-throughput sequencing technology was performed after the treatment of human ovarian cancer cells A2780 with TMPyP4 and H2O2 purchased from Sigma-Aldrich, to explore the expression of genes related to cell proliferation, DNA damage and ROS levels of ovarian cancer cells A2780 after the treatment of TMPyP4 and H2O2, and to find an interesting target pathway,HIF-1 signaling pathway. Overall design: In dark conditions, human ovarian cancer cells A2780 treated with TMPyP4 at the concentration of 5 µM for 72 hours is the TMPyP4 group, A2780 incubated for 46 hours and then treated with H2O2at the concentration of 200 µM for 2 hours and incubated for another 24 hours is the H2O2 group, A2780 treated with TMPyP4 at the concentration of 5 µM for 46 hours and then treated with H2O2at the concentration of 200 µM for 2 hours and incubated with TMPyP4 for another 24 hours is the co-treatment of TMPyP4 and H2O2 group. The experimental design of light conditions is same as dark conditions, with the exception of a 3-minute exposure to 600 nm white light at 48 hours.
