Dataset for "Systematic mapping of orthogonality and domain-swap permissiveness across LysR-type transcriptional biosensors" (Demeester et al. 2026)

This repository contains the underlying data and analysis code for the study "Systematic mapping of orthogonality and domain-swap permissiveness across LysR-type transcriptional biosensors" (Demeester, Declerck, De Mey & De Paepe, Ghent University). The work characterises a panel of LysR-type transcriptional regulator (LTTR) biosensors in Escherichia coli using plate-reader assays (OD600 and mKate2 fluorescence, n = 4 biological replicates), across four experiment blocks: a functionality screen of 17 candidate LTTRs, a pairwise orthogonality (cross-talk) matrix, a domain-swapping campaign generating chimeric regulators, and an orthogonality-transfer case study. The deposit includes the raw per-well measurement tables and plate maps, the per-figure source data (main Figures 2, 3, 5 and 6), and the Jupyter notebooks and helper functions that reproduce the figures, statistics and tables.
