Antioxidant effect of Cymbopogon citratus and Hypericum perforatum in boar semen like a new alternative for it conservation

Boar semen is highly susceptible to lipid peroxidation process, sperm capacitation and early acrosomal reaction, which is why the evaluation of natural extracts of Cymbopogon citratus and Hypericum perforatum is proposed as a new alternative for seminal preservation and delaying spermatozoa damage for a period of time of 72 hours. In the present research, three different solutions (0.250, 0.125 and 0.0625 µl) of each extract were mix with fresh boar semen. The extracts were characterized by DPPH* and HPLC for the determination of antioxidants and total sugars. Seminal viability was analyzed using a Seminal Quality System. The results were compared with a short-term extender, obtaining as a result for Cymbopogon citratus a viability of 66% vs. 63% and, for Hypericum perforatum 82% vs. 63%, in a period of three days. By using these natural extracts, it was possible to maintain sperm viability above 60% for 72 hours in storage.

公猪精液极易受到脂质过氧化(lipid peroxidation)、精子获能(sperm capacitation)及早期顶体反应(early acrosomal reaction)过程的影响而发生损伤,因此本研究提出将柠檬草(Cymbopogon citratus)与贯叶连翘(Hypericum perforatum)的天然提取物作为精液保存的新型替代方案,以延缓精子损伤,使储存时长可达72小时。本研究将每种提取物的三种不同剂量(0.250、0.125及0.0625 µl)分别与新鲜公猪精液混合。采用DPPH(DPPH*)自由基清除法与高效液相色谱(HPLC)对提取物的抗氧化成分及总糖含量进行表征分析。采用精液质量分析系统(Seminal Quality System)检测精子活力。将实验结果与短期稀释液(short-term extender)的效果进行对比,结果显示:在为期三天的储存周期内,柠檬草提取物组的精子活力为66%,对照组为63%;贯叶连翘提取物组的精子活力为82%,对照组为63%。使用上述天然提取物后,可使储存72小时后的精子活力维持在60%以上。
