Leptinotarsa decemlineata Raw sequence reads

This Bioproject contains raw reads used for bulk segregant analysis of pooled samples of Leptinotarsa decemlineata used to identify genomic regions involved in resistance against the insecticide imidacloprid.The Colorado potato beetle (CPB, Leptinotarsa decemlineata) is a devastating pest of potato and is resistant to many insecticides including imidacloprid. The broader aim of our study was to identify genes associated with imidacloprid resistance in CPB. We crossed five strains of CPB collected from different parts of Europe to generate a mapping population. Then, we performed toxicity assays on the F4 2nd instar larvae with either a high (250ppm) or a low (1ppm) concentration of imidacloprid. The larvae that survived the high concentration assay (n=51) formed the 'resistant' bulk and the larvae that died in the low concentration (n=66) formed the 'susceptible' bulk. DNA was isolated from the pools and whole genome sequencing was performed. Bulk Segregant Analysis was then performed with 'QTLseqr' following the G' analysis to identify genomic regions corresponding to resistance.
