Purpose: to determine changes in RNA expression in WT vs BST2 KO mice following 24 hour treatment of IFNg. We were particularly interested in performing pathway analysis using GSEA. Overall design: HSC (Lineage- c-Kit+ CD150+ CD48- CD34-) were sorted using a Sony cell sorter, SH800. 5,000-11,000 HSCs were isolated from each mouse and RNA was isolated using RNeasy kit (Qiagen, Cat#74004). RNA-sequencing library was generated using SMARTer® Stranded Total RNA-Seq Kit V3 (Takara Bio USA). The sequencing was performed using Illumina NovaSeq SP with a paired-end sequencing length of 10bp.
