ChIP-seq was carried for zfp148 and zfp281 in human K562 cells to test the hypothesis that these two transcription factor family members play overlapping roles in erythropoiesis. Overall design: Because of concern for possible cross-reactivity of antibodies against zfp148 and zfp281, K562 cells stably expressing the bacterial biotin ligase Bir A were stably transfected with recombinant forms or either zfp148 and zfp281 containing a 23 amino acid amino terminal BirA recognition/biotin accepter motif ("bio-zfp148" and "bio-zfp281"). Clones were selected that expressed the recombinant proteins at close to the same levels as the endogenous proteins. The parental K562 cells expressing BirA alone were included as controls. The cells were treated with 50 mM hemin (final conc) for 48 hrs to induce erythroid maturation, and then streptavidin-based ChIP-seq was performed.
