本研究通过基于细菌人工染色体(bacterial artificial chromosome)构建的转基因小鼠模型(Ube3aOE),探究Ube3a基因剂量过高对神经发育的影响;该模型可精准复刻15q11-q13重复综合征(Dup15q)中最常见的Ube3a拷贝数增加现象。在此基础上,本研究利用野生型与Ube3aOE小鼠皮层及海马组织的RNA-seq数据开展基因表达谱分析,并针对5只野生型小鼠与5只Ube3aOE小鼠的皮层及海马脑组织RNA-seq数据完成对比基因表达谱分析。
