Gene expression profile at single cell level from Glycine max nodules and roots.

The nuclei of Glycine max root seedlings (6 days-old) and nodules (28 days-old) were isolated and the library consstruction was performed applying 10x Genomics tecnology. The nuclei of the 6 days-old roots of Glycine max seedlings grown on B&D-N media and growth chambers in dark. For nodule sample, the 3-day-old seedlings were germinated between moist Whatman filter paper and were inoculated with 1 mL of B. japonicum suspension (OD600=0.1) on transferred 3‐day‐old seedlings. The nodules were collected after 28 days. For the nuclei isolation was used mechanical and chemical steps and the libraries were made following 10X genomics recommendations. The pre-processing and processing of the datasets were performed with Cell Ranger, and Seurat.
