在微流控芯片中,通过共培养人脑微血管内皮细胞、星形胶质细胞和小胶质细胞来构建体外血脑屏障模型。该模型可部分模拟体内血脑屏障的结构与功能,并再现脑组织微环境特征。将该模型用于恶性黑色素瘤来源外泌体暴露研究,可观察到血脑屏障内皮损伤与胶质瘤激活的过程,并研究其潜在机制。

An in vitro blood-brain barrier (BBB) model is established by co-culturing human brain microvascular endothelial cells, astrocytes, and microglia in a microfluidic chip. This model can partially simulate the structure and function of the in vivo blood-brain barrier, and recapitulate the features of the brain tissue microenvironment. When applied to studies on exposure to exosomes derived from malignant melanoma, this model allows observation of the processes of BBB endothelial injury and glioma activation, and facilitates investigation of their underlying mechanisms.
