KIF18A inhibition causes hyper-oscillation of mitotic chromosomes and spindle assembly checkpoint activation at attached chromosomes. In a subset of cancers, these phenotypes drive dramatic mitotic delays, division errors, and cell death. In other cancers and non-transformed cells however, KIF18A inhibition is well-tolerated. We were interested in whether these KIF18A agnostic cell lines were still experiencing chromosome mis-segregation errors despite continued growth. Overall design: Three KIF18A agnostic cell lines (RPE1, MCF10A, and HCT116) were treated with KIF18A inhibitor for 7 days, then their DNA content was compared to the DNA content of untreated cells using scDNAseq.
