The mutant siaux1-1 and as well as wildtype Yugu1 were grown in soil mixture containing packaged potting soil, peat moss and sand (2:2:1, v/v) in a growth chamber (10 hours with 30? in Day to 14 hours with 26? as night). Totally 10 tissue samples were collected for RNA sequencing for the mutant and wildtype plant, including leaf and root tissues from seedling plants when growing out one fully expanded leaf (1 leaf stage), leaf and root tissues at 3 leaf stage, and young panicle at heading stage. All tissues were immediately frozen in liquid nitrogen and stored at -80?°C. RNA extractions were preformed using Trizol (Ambion, Thermo Scientific Inc., USA) method with three independent replications for each tissue. RNA samples were sent to Beijing Berry Genomic (Company Limited, Beijing, China) for transcriptome sequencing. Thirty cDNA libraries were constructed and sequenced on Illumina HiSeq X ten platform in 150-bp paired-end mode (Illumina Inc., USA).
