This study is to investigate the potential impact of an lncRNA NR_126553 (Nostril) in murine intestinal epithelial cells (IEC4.1 cells) in response to IFN-gamma protein (IFN?) stimulation. NR_126553 (Nostril) was knocked down by using a pool of gene specific siRNA (SiNostril). Cells treated with scramble non-specific siRNA were used as control (siNegative control). After siRNA transfection 24h, cells were treated with or without mouse IFN? (1ng/ml for 4hrs). Then total RNA was collected for sequencing via DNBSEQ platform to obtain a comprehensive view of the transcriptome. Overall design: IEC 4.1 cells were transfected with the siRNAs (or SiNegative control) for 24 hours to allow gene knockdown or other desired effects. Four groups were included in this set up, Group A: Non-specific scrambled siRNA control treatment. Group B: Non-specific scrambled siRNA control treatment + IFN? stimulation. Group C: Double-strand RNA encoding siNR_126553 (SiNostril) treatment. Group D: Double-strand RNA encoding siNR_126553 ( (SiNostril) +IFN? stimulation. Total RNAs were prepared with the RNeasy Mini kit (Qiagen) according to the manufacturer's instruction.
