Antibody glycosylation plays a crucial role in the humoral immune response by regulating effector functions and influencing binding affinity to immune cell receptors. While antibody glycosylation has been studied extensively for its potential to predict disease status, previous studies have focused mainly on the immunoglobulin G (IgG) isotype due to analytical challenges associated with other isotypes. Thus, developing a sensitive and accurate analytical platform is necessary to characterize antibody glycosylation across multiple isotypes. Our platform targets 98 glycopeptides across IgG, IgA, and IgM isotypes, as well as eight surrogate peptides representing total IgG, four IgG classes, two IgA classes, and IgM. The developed method is sensitive and accurate for Ig glycosylation profiling. It can be adopted for clinical investigations, particularly for detailed humoral immune profiling.

抗体糖基化通过调控效应功能、影响与免疫细胞受体的结合亲和力,在体液免疫应答中发挥关键作用。尽管学界已围绕抗体糖基化预测疾病状态的潜力开展了大量研究,但由于其他免疫球蛋白亚型存在分析检测难点,既往研究多聚焦于免疫球蛋白G(immunoglobulin G,IgG)亚型。因此,亟需开发一款灵敏精准的分析平台,以表征多种免疫球蛋白亚型的抗体糖基化特征。本研究开发的分析平台可靶向覆盖IgG、IgA、IgM三种亚型共98条糖肽,同时包含8条替代肽,分别对应总IgG、4个IgG亚类、2个IgA亚类及IgM。该方法可实现免疫球蛋白糖基化谱的灵敏精准分析,可应用于临床研究,尤其适用于精细化体液免疫谱分析。
