To assess the genetic stability of a CRISPRi-Cas12m biosecure design, we conducted a long-term growth experiment, passaging the strain daily under permissive conditions for 21 days. Whole-genome sequencing was performed on both the original and evolved strains to identify any mutations that may have occurred within the three Cas12m insertions in the E. coli chromosome.
