通过纳氏试剂分光光度法检测采样点的氨氮(NH3-N)值,反映水体中氨氮的含量,用于监测废水中的氮污染程度,为定海区千岛街道河道水的达标排放等提供数据支持。水样采集:从各个检测点位中采得水样。 水样保存:水样采集在聚乙烯瓶或玻璃瓶内,并应尽快分析,必要时可加硫酸将水样酸化至pH<2,于2- 5°C下存放。酸化样品应注意防止吸收空气中的氮而遭致污染。 水样预处理:水样带色或浑浊以及含其它一些干扰物质,影响氨氮的测定。为此,在分析时需做适当的预处理。对较清洁的水,可采用絮凝沉淀法,对污染严重的水或工业废水则以蒸馏法使之消除干扰。 检测方法:以游离态的氨或铵离子等形式存在的氨氮与纳氏试剂反应生成淡红棕色络合物,该络合物的吸光度与氨氮含量成正比,于波长420nm处测量吸光度。 结果分析:水中氨氮的浓度按公式(1)计算: ρN=(AS-Ab-a)/(b×V) 式中: ρN——水样中氨氮的质量浓度,mg/L,以氮计; AS——水样的吸光度; Ab——空白试验的吸光度; a——校准曲线的截距; b——校准曲线的斜率; V——试样体积,mL。
The ammonia nitrogen (NH3-N) concentration at sampling sites was detected via Nessler's reagent spectrophotometry to reflect the ammonia nitrogen content in water bodies, monitor the nitrogen pollution level of wastewater, and provide data support for compliant discharge of river water in Qiandao Subdistrict, Dinghai District, etc.
Water sample collection: Water samples were collected from all detection points.
Water sample preservation: Water samples should be collected in polyethylene or glass bottles and analyzed as soon as possible. If necessary, add sulfuric acid to acidify the samples to pH < 2, and store them at 2–5 °C. Attention shall be paid to preventing acidified samples from absorbing nitrogen in the air to avoid contamination.
Water sample pre-treatment: Colored, turbid water samples or those containing other interfering substances may affect the determination of ammonia nitrogen. Therefore, appropriate pre-treatment is required prior to analysis. For relatively clean water, the flocculation precipitation method can be used; for severely polluted water or industrial wastewater, the distillation method is adopted to eliminate interferences.
Detection method: Ammonia nitrogen existing in the form of free ammonia or ammonium ions reacts with Nessler's reagent to generate a light reddish-brown complex. The absorbance of this complex is proportional to the ammonia nitrogen content, and the absorbance is measured at a wavelength of 420 nm.
Result analysis: The concentration of ammonia nitrogen in water is calculated using formula (1):
ρN = (AS - Ab - a) / (b × V)
Where:
ρN — Mass concentration of ammonia nitrogen in water samples, mg/L, expressed in terms of nitrogen;
AS — Absorbance of the water sample;
Ab — Absorbance of the blank test;
a — Intercept of the calibration curve;
b — Slope of the calibration curve;
V — Volume of the test sample, mL.
