Next generation RNA sequencing was employed to determine the acute and sub-chronic impact of peristaltic pulse external pneumatic compression (PEPC) of different target inflation pressures on global gene expression in human vastus lateralis skeletal muscle biopsy samples. Methods: Eighteen (N=18) male participants were randomly assigned to one of three groups: 1) sham (n=6), 2) EPC at 30-40 mmHg (LP-EPC; n=6) and 3) EPC at 70-80 mmHg (MP-EPC; n=6). 1-h treatment with sham/EPC occurred for 7 consecutive days. Vastus lateralis skeletal muscle biopsies were performed at baseline (before first treatment; PRE), 1-h following the first treatment (POST1), and 24-h following the last (7th) treatment (POST2). Changes from PRE in gene expression were analyzed via paired comparisons within each group. Genes were filtered to include only those that had an RPKM > 1.0, a fold-change of > 1.5 and a paired t-test value of <0.01. Results: For the sham condition, 2 genes at POST1 and 1 gene at POST2 were significantly altered. For the LP-EPC condition, 9 genes were upregulated and 0 genes were downregulated at POST1 while 39 genes were upregulated and 1 gene downregulated at POST2. For the MP-EPC condition, 2 genes were significantly upregulated and 21 genes were downregulated at POST1 and 0 genes were altered at POST2. Conclusions: Both LP-EPC and MP-EPC acutely alter skeletal muscle gene expression though only LP-EPC appeared to affect gene expression with sub-chronic application. Moreover, the transcriptome response to EPC demonstrated marked heterogeneity (i.e., genes and directionality) with different target inflation pressures.
本研究采用下一代RNA测序技术,探究不同目标充气压力的蠕动脉冲式外部气压加压(peristaltic pulse external pneumatic compression, PEPC)对人类股外侧肌骨骼肌活检样本中全基因组基因表达的急性及亚慢性影响。
方法:18名(N=18)男性受试者被随机分为三组:1)假处理组(sham组,n=6);2)30~40 mmHg气压加压组(LP-EPC组,n=6);3)70~80 mmHg气压加压组(MP-EPC组,n=6)。所有受试者接受每日1小时的假处理或气压加压处理,连续干预7天。分别于基线期(首次处理前,记为PRE)、首次处理后1小时(记为POST1)以及末次(第7次)处理后24小时(记为POST2)采集股外侧肌骨骼肌活检样本。采用组内配对比较分析各组样本相较于PRE的基因表达变化。对基因进行筛选,仅保留每百万映射reads的外显子片段数(Reads Per Kilobase of transcript per Million mapped reads, RPKM)>1.0、倍数变化(fold-change)>1.5且配对t检验P值<0.01的基因。
结果:假处理组中,POST1时点有2个基因、POST2时点有1个基因的表达发生显著改变。LP-EPC组中,POST1时点有9个基因上调、0个基因下调;POST2时点则有39个基因上调、1个基因下调。MP-EPC组中,POST1时点有2个基因显著上调、21个基因下调,而POST2时点无基因表达发生显著改变。
结论:LP-EPC与MP-EPC均可急性改变骨骼肌基因表达,但仅LP-EPC在亚慢性干预下可对基因表达产生影响。此外,不同目标充气压力下,骨骼肌转录组对EPC的响应存在显著异质性(即涉及基因种类及表达调控方向均存在差异)。
