The transcriptomes of single cells from human bone marrow were investigated at single cell level, with the aim to reconstruct the development of human dendritic cell lineages. Overall design: 399 human bone marrow mononuclear cells were isolated into 96 well plates via fluorescence-activated cell sorting (FACS). CD34+ cells from the following phenotypic spaces were sequnced at single cell level: CD38-CD45RA-hematopoietic stem cells (HSC) and multipotent progenitors (MPP), CD38+CD45RA- common myeloid progenitors (CMP), CD38-CD45RA+ lymphoid-primed multipotent progenitors (LMPP) and CD38+CD45RA+ granulocyte-macrophage progenitors (GMP).
