deep mutational scanning of the SARS-CoV-2 RBD using yeast display of barcoded libraries

We performed deep mutational scanning of the SARS-CoV-2 spike receptor-binding domain (RBD) from strain Wuhan-Hu-1 using yeast display to measure the affects of all amino-acid mutations on phenotypes such as ACE2 binding affinity and RBD expression. We then used this same library to perform mutational antigenic profiling to map RBD mutations that escape antibody binding.
