Purpose: The goal of this study to examine mRNA transcriptomic changes in reward-related brain regions of subjects with alcohol use disorder.
Methods: Total RNAs were extracted from postmortem caudate nucleus of 12 AUD and 12 control subjects. rRNA depletion RNA sequencing was performed and the sequence reads were processed using the bulk RNA-seq processing pipeline Pipeliner workflow (Federico et al. Front Genet 2019; 10, 614). AUD-associated mRNA transcriptomic changes were analyzed by the Limma-Voom method.
Results: Differentially expressed mRNAs (absolute FC>2.0 & P<0.05) were identified in postmortem caudate nucleus of subjects with alcohol use disorder (AUD).
Chronic alcohol consumption may alter mRNA transcriptome profiles in reward-related brain regions, resulting in alcohol-induced neuroadaptations.
研究目的:本研究旨在探究酒精使用障碍(Alcohol Use Disorder, AUD)患者奖赏相关脑区的mRNA转录组变化。
研究方法:从12名酒精使用障碍患者及12名对照受试者的死后尾状核中提取总RNA。采用核糖体RNA(ribosomal RNA, rRNA)去除法进行RNA测序,并使用批量RNA测序分析流程Pipeliner工作流(Federico等,Front Genet 2019; 10, 614)对测序读段进行处理。通过Limma-Voom方法分析与AUD相关的mRNA转录组差异变化。
研究结果:在酒精使用障碍患者的死后尾状核中,鉴定出绝对FC>2.0且P<0.05的差异表达mRNA。
慢性酒精摄入可改变奖赏相关脑区的mRNA转录组谱,进而引发酒精诱导的神经适应性改变。
