STAT6 turnover

Decay of Stat6 activation in Ttc7-fsn B cells. Purified splenic B cells were stimulated with IL-4 for 30 minutes, washed 3X and allowed to rest for 15 or 30 minutes, then stained for Stat6 (red) and the nucleus (green) and imaged with confocal microscopy using a 60x objective. Co-localization of Stat6 with nucleus, denoting activation, is yellow. Ttc7fsn cells show a similar trend to WT cells at both time points, indicating they have the inherent ability to downregulate Stat6 activation when IL-4 stimulus is removed.

Ttc7-fsn B细胞中Stat6激活的衰减过程。本实验将纯化的脾脏B细胞用白细胞介素4（IL-4）刺激30分钟，经3次洗涤后静置15或30分钟；随后对Stat6（红色荧光标记）与细胞核（绿色荧光标记）进行染色，并采用60倍物镜通过共聚焦显微镜成像。Stat6与细胞核的共定位代表激活状态，此时呈现黄色。Ttc7fsn细胞在两个观测时间点均表现出与野生型（Wild Type, WT）细胞相似的变化趋势，这表明在移除IL-4刺激后，Ttc7fsn细胞具备下调Stat6激活的内在能力。
