This project is designed to analyse the inter-relationships of DNA methylation and hydroxymethylation, chromatin structure and MeCP2. It mainly utilises a human neuronal progenitor cell line called LUHMES. This cell line grows rapidly in the presence of exogenous Myc, but when this transgene is inactivated, the progenitors differentiate in culture into dopaminergic neurons. We will analyse DNA methylation and hydroxymethylation by bisulfite sequencing chromatin structure using various antihistone modifcation antibodies, and MeCP2 binding also by ChIP.
