OSSL_325096 is a compound that acts as p97/VCP inhibitor and induces apoptosis in myeloma cells. Therefore, we treated KMS12PE and KMS11 myeloma cell lines with OSSL_325096 or DMSO and compare gene expression levels to elucidate the mechanisms of apoptosis. We treated KMS12PE and KMS11 myeloma cell lines with 1 μM OSSL_325096 or DMSO. RNA was extracted and purified using Trizol reagent (Thermo Fisher Scientific) and an RNeasy Mini Kit (Qiagen). Primary RNA-sequencing was performed by Novogene (Beijing, China). Raw sequence data was mapped to hg19 and expression values were determined as RPKM using Biomedical Genomics Workbench 5 software (QIAGEN Bioinformatics, Venlo, Netherlands). Then gene expression levels in two myeloma cell lines were compared using Biomedical Genomics Workbench 5 software.
