To resolve the molecular mechanism of Hey2 in cardiac energy metabolism, we performed CUT&Tag-seq (cleavage and under targets tagmentation sequencing) in ventricular cells isolated from myocardium specific hey2- overexpressing (hey2-mOE) zebrafish hearts and control zebrafish hearts. To further decipher the impact of HEY2 in mammalian cardiomyocytes and to evaluate histone acetylation modifications, we performed Flag-HEY2, H3K9ac and H3K27ac CUT&Tag-seq on NRVMs infected with adenovirus containing Flag-Hey2 (Adv-Hey2) or vectors (Adv-Ctrl).
