The study aims to explore the protective effects of oridonin against hepatic ischemia–reperfusion injury (IRI) and its underlying mechanisms. Hepatic IRI is a significant clinical issue that can lead to severe liver dysfunction. Oridonin (Ori), a natural diterpenoid compound, has shown promising therapeutic potential. In an in vivo model of hepatic IRI, oridonin (5 mg/kg) treatment significantly reduced serum levels of liver injury markers. In this study, we comprehensively investigated the pharmacological effects of oridonin in the treatment of liver IRI and clarified the potential molecular mechanisms. Overall design: RNA sequencing was performed on liver tissue from wild-type C57BL/6 mice subjected to ischemia–reperfusion injury, with and without oridonin.
