Monosomic Alien chromosome addition lines (MAALs) have established in a variety of crops by adding a single chromosome of donor species to introduce valuable traits. Cytological analyses indicated that genome rearrangements were induced during establishment of MAALs, however, precise genome structure of MAALs are yet to be determined. Single-molecule real-time (SMRT) sequencer, which enables to sequence long DNA fragments, is a powerful tool to assemble accurate and contiguous genomes. Yasui and Iwata have established MAALs in Oryza sativa cultivar Nipponbare carrying a single chromosome of O. punctata, distantly related species of O. sativa. In order to understand the genome structure of the MAALs, we have sequenced the genomes of the MAALs and their parental lines with PromethION, a SMRT sequencer developed by Oxford Nanopore Technologies.
