Mesenchymal stem/stromal cells (MSCs) demonstrate great therapeutic potential in regenerative medicine. However, the differences between the transcriptome of MSCs from umbilical cord tissue and in vitro cell culture at single-cell level remain elusive. We obtained a large-scale single-cell transcriptome of total 19221 single cells from human umbilical cord Wharton's jelly tissue and the matched culture-expanded MSCs from three donors to provide a high quality transcriptomic profiling dataset at single-cell resolution, which is a valuable resource for dissecting the MSCs heterogeneity, cell fate decision in native tissue niche and in vitro culture environment for specific lineages.

间充质干/基质细胞(Mesenchymal stem/stromal cells, MSCs)在再生医学领域展现出巨大的治疗应用潜力。然而,脐带组织来源的间充质干/基质细胞与体外培养细胞的转录组在单细胞层面的差异仍未明确。本研究从3名供者的人脐带沃顿胶(Wharton's jelly)组织及配对的体外扩增培养的间充质干/基质细胞中,共获取了19221个单细胞的大规模单细胞转录组数据,构建了具备高单细胞分辨率的高质量转录组图谱数据集。该数据集可为解析间充质干/基质细胞的异质性、天然组织微环境中的细胞命运决定,以及特定细胞谱系的体外培养环境调控机制提供宝贵的研究资源。
