RNA-seq of Bone Marrow Mesenchymal Stem Cells under Cyclic Mechanical Stretch

Mesenchymal stem cells (MSCs) are highly sensitive to mechanical stimuli, which play a crucial role in their differentiation and function. This study aims to investigate the global transcriptional changes in bone marrow-derived MSCs (BMSCs) in response to cyclic mechanical stretch. BMSCs were subjected to 15% deformation with a 5-second dwell time at maximum deformation using a self-developed spherical gradient stretching device for 1440 cycles. Total RNA was extracted from both stretched and static control groups (n=3 biological replicates per group). RNA-seq libraries were prepared with poly-A selection and sequenced on the Illumina NovaSeq 6000 platform, generating paired-end reads. The resulting data provides a comprehensive resource for understanding the mechanotransduction mechanisms in BMSCs and identifying novel mechanosensitive genes and pathways involved in osteogenesis, chondrogenesis, or other lineage commitments.
