Role of Wnt signalling in the self-renewal and differentiation capacities of primary hepatoblast organoids.

Primary hepatoblast organoids (HBOs) are grown and passaged in medium that contains both Wnt3a and R-spondin conditioned media (HBO complete medium). It is hypothesised that these molecules that are part of the WNT signalling pathway may be contributing to the HBOs self-renewal properties. Overall design: To investigate the function of Wnt3a and R-spondin on the self-renewal as well as differentiation capacity of the HBOs the removal of the signalling molecules from the HBO complete medium was tested, separately as well as combined.
