Short capped RNAs

In eukaryotes, capped RNAs include long transcripts such as messenger RNAs and long non-coding RNAs, as well as shorter transcripts such as spliceosomal RNAs, small nucleolar RNAs, and enhancer RNAs. Long capped transcripts can be profiled using Cap Analysis Gene Expression (CAGE) sequencing and other methods. We developed a novel method for sequencing library preparation that combines 5' cap detection with RNA size selection to specifically select for short capped RNAs. We applied this method to RNA of a differentiation time course of the THP-1 leukemia cell line, performed deep sequencing, and systematically compared the landscape of short capped RNAs to that of long capped RNAs.
