DNA from iPSCs was sheared using a Covaris S2 Focused-ultrasonicator:130-ml samples in microTUBEs were subjected to two 60-s cycles of 10%duty cycle, intensity of 4, and 200 cycles per burst in frequency sweepingmode. DNA Chip (DNA 1000 Kit; Agilent Technologies) analysis using an Agi-lent 2100 Bioanalyzer indicated an average fragment size of 400 bp. Librarypreparation was performed using an NEBNext Ultra DNA Library Prep Kit forIllumina (NEB #E7370), and the ?nal library concentration was determined byqPCR using a KAPA Illumina Library Quanti?cation Kit (KK4835; KAPA Bio-systems) and an Applied Biosystems ViiA 7 Real-Time PCR System (LifeTechnologies).Sequencing clusters were produced on the ?ow cell using an IlluminaTruSeq PE Cluster Kit v.3-cBot-HS (PE-401-3001) and an Illumina cBot.WGS was performed using an Illumina TruSeq SBS Kit v.3-HS-200 cycles(FC-401-3001) and an Illumina HiSeq 2500 upgrade to generate 2 3 100single-index paired-end reads for bioinformatic analysis.
