Nucleosides and nucleotide analogues are synthetic. Substances that have undergone certain chemical modifications can mimic nucleosides and nucleotides in the body and are involved in DNA or RNA synthesis. Because they don't work properly, they can stop cell division or viral replication. In addition to being involved in nucleic acid composition, nucleoside and nucleotide analogues can act on some important enzymes in the body, inhibiting enzyme activity, such as human and viral polymerases (DNA-dependent DNA polymerase, RNA-dependent DNA polymerase, or RNA-dependent RNA polymerase), kinases, nucleotide reductases, DNA methyltransferases, purines and pyrimidine nucleoside phosphorylases, and thymidine synthases. These mechanisms of action of nucleosides and nucleotide analogues have potential applications in inhibiting cancer cell growth, viral replication, and treating other indications. In order to integrate the data resources and results of numerous nucleoside drug studies, facilitate comparative studies, and carry out literature review and information extraction analysis, a nucleoside drug knowledge base containing 553 pieces of information was constructed to assist nucleoside drugs in the treatment of cancer and viruses. The database covers drug name, structural formula, structure, catalog name, CAS number, relative molecular weight, biological activity, target, pathway, solubility, concentration, reference title, and corresponding website sources. This API supports query functions such as CAS drug registration number and structure. Therefore, the nucleoside drug database will help to find new indications for cancer treatment from the idea of "old drugs for new purposes", in addition to the efficacy of the nucleoside drugs themselves.
