<b>FIG S2 </b><b>Transition metals (Cu, Zn, Ni) do not affect HOSCN stability. </b>Reactions (1 ml) containing 500 µM HOSCN and either no metal or 100 µM of CuCl₂, ZnSO₄, or NiSO₄ in 10 mM HEPES-KOH buffer (pH 7.4) were incubated for 20 min at 37°C. At 5-min intervals, 20 µl aliquots were removed and assayed for HOSCN content using 2-nitro-5-thiobenzoic acid (TNB)(n=3 experimental replicates; error bars of 1 standard deviation).

反应体系总体积为1 ml，包含500 µM次硫氰酸（HOSCN），分别设置无金属离子对照组、100 µM氯化铜（CuCl₂）组、100 µM硫酸锌（ZnSO₄）组及100 µM硫酸镍（NiSO₄）组，所有体系均置于10 mM HEPES-KOH缓冲液（pH 7.4）中，于37℃孵育20 min。每间隔5 min取20 µl等分试样，采用2-硝基-5-硫代苯甲酸（2-nitro-5-thiobenzoic acid，TNB）法检测次硫氰酸（HOSCN）含量，实验重复3次，误差棒代表1倍标准差。
