For genome sequencing, young leaves from a 4-year-old E. petiolatus plant were collected in Menglun, Xishuangbanna, China. High-molecular-weight DNA was extracted using a modified CTAB method, and SMRTbell libraries were prepared according to the PacBio 15-kb protocol. Sequencing was carried out using circular consensus sequencing (CCS) on a PacBio Revio platform, generating HiFi long reads for genome survey analysis and long-read assembly. Hi-C libraries were prepared from over 2 g of young leaves through chromatin extraction, digestion, ligation, and DNA purification, and sequenced on an Illumina NovaSeq 6000 platform to produce paired-end reads for scaffolding. For transcriptomic analysis, total RNA was isolated from the fruit, stem, and leaves of the same plant, and RNA sequencing (RNA-seq) libraries were prepared using the TruSeq Stranded mRNA-Seq protocol. Sequencing was also conducted on the Illumina NovaSeq 6000 platform. The RNA-seq reads were used for genome quality assessment and transcriptome-based gene annotation. In total, the sequencing effort yielded 11.23 Gb of HiFi reads, 42.45 Gb of Hi-C reads, and 17.59 Gb of RNA-seq reads.
