Identification and experimental validation of mitochondrial metabolism-related biomarkers in chronic rhinosinusitis with nasal polyps

In this paper, we show that mitochondrial metabolism-related genes (MM-RGs) play a critical role in the pathogenesis of chronic rhinosinusitis with nasal polyps (CRSwNP). This is significant because, by integrating transcriptomic and single-cell RNA sequencing data, we identified five key biomarkers—CYP2J2, CYBB, ALOX5, PLB1, and PLA2G2A—which are enriched in immune-related pathways and strongly associated with M2 macrophage and plasma cell infiltration. Furthermore, our predictive nomogram and identification of epithelial and T cells as primary mediators, alongside validation through RT-qPCR, highlight novel diagnostic and therapeutic avenues for CRSwNP.
本研究证实,线粒体代谢相关基因(mitochondrial metabolism-related genes)在伴鼻息肉的慢性鼻-鼻窦炎(chronic rhinosinusitis with nasal polyps, CRSwNP)的发病机制中发挥关键作用。本研究通过整合转录组学与单细胞RNA测序(single-cell RNA sequencing)数据,鉴定出CYP2J2、CYBB、ALOX5、PLB1及PLA2G2A这5种关键生物标志物,上述标志物显著富集于免疫相关通路,且与M2型巨噬细胞及浆细胞浸润密切相关。此外,本研究构建的预测列线图、明确上皮细胞与T细胞为主要介导细胞,以及通过实时定量聚合酶链反应(RT-qPCR)开展的验证工作,为伴鼻息肉的慢性鼻-鼻窦炎提供了全新的诊断与治疗思路。
