Cellular barcodes in human fibroblasts and human induced pluripotent stem cells extracted from singe-cell copy DNA

Single cell RNA sequencing (10x Genomics single-cell RNA-seq kit v3) of barcoded human fibroblast-like cells (hiF-T cells) and human induced pluripotent stem cells (hiPS cells) derived from hiF-T cells. hiF-T cells were transduced with barcode libraries consisting of semi-random 100-mers in the 3' UTR of GFP and permitted to undergo several cell divisions before being split and seeded for parallel reprogrammings. Both barcoded hiF-T cells and hiPS cells were FACS sorted for GFP positivity to enrich for barcode-containing cells before performing single-cell RNA-sequencing. Barcodes were recovered from GFP transcripts via a separate "side reaction" and sequencing run on cDNA. See Jain et al. citation for additional details.
