Y. pestis strain 201was cultivated under various conditions to achieve the greatest diversity of mRNA transcripts, including 26 degree in TMH, 37 degree in TMH without calcium (pH6.0), 37 degree in TMH with 2.5% NaCl, 4 degree in TMH, 42 degree in TMH in an incubator shaking at 200 rpm overnight. Equal amount bacterial cells grown under the aforementioned conditions were mixed together and the total RNA was then isolated. One cDNA library was constructed for RNA-seq analysis.
