Figure 3: Gene expression analysis of mercury compounds in medaka embryos. Medaka embryos were sampled from control, 0.1 μM MeHg, 1 μM HgCl2, 10 μM α-HgS, and 10 μM β-HgS treatment groups (n=3, 15 embryos pooled per replicate) at 3 days post-exposure. Total RNA was extracted and subjected to RT-PCR analysis for metallothionein (<i>MT</i>) and heme oxygenase-1 (<i>Ho-1</i>) gene expression using β-actin expression as the reference. Data are mean ± SD. *Significantly different from controls with p < 0.05.
图3:青鳉胚胎内汞化合物的基因表达分析。本实验设置对照组、0.1μM甲基汞(MeHg)、1μM氯化汞(HgCl₂)、10μM α-硫化汞(α-HgS)及10μM β-硫化汞(β-HgS)处理组,于暴露后3天采集胚胎样本(每组生物学重复数n=3,每个重复混合15枚胚胎)。提取总RNA后,以β-肌动蛋白(β-actin)的表达作为内参基因,通过逆转录聚合酶链反应(RT-PCR)检测金属硫蛋白(MT)与血红素加氧酶-1(Ho-1)的基因表达水平。实验数据以平均值±标准差(SD)呈现,*标记表示与对照组相比具有统计学显著性差异(p<0.05)。
