Lipofectamine transfection of SARS-CoV-2 replicon DNA leads to GFP expression that is dependent on DNA linearization but not T7 RNAP and is remdesivir-insensitive.</b> HEK293T transduced with LPCX(AB) (A) or with LPCX(AB)-T7RNAP (B) were then transfected with pSMART-BAC-T7-scv2 DNA, linearized or not, and in the absence or presence of remdesivir, exactly as in Fig. 2, except that transfections were performed using Lipofectamine 3000. Cells were analyzed by flow cytometry two days later.

<b>图3 Lipofectamine转染严重急性呼吸综合征冠状病毒2型（SARS-CoV-2）复制子DNA可诱导绿色荧光蛋白（GFP）表达，该表达依赖于DNA的线性化过程，不依赖T7 RNA聚合酶（T7 RNAP），且对瑞德西韦不敏感。</b> 将分别经LPCX(AB)（对应图A组）或LPCX(AB)-T7RNAP（对应图B组）转导的HEK293T细胞，使用pSMART-BAC-T7-scv2 DNA（已线性化或未线性化）进行转染，实验设置添加瑞德西韦与不添加瑞德西韦两组，实验流程与图2完全一致，仅转染步骤改用Lipofectamine 3000完成。转染两天后，通过流式细胞术对细胞进行分析。
