PetB metabarcoding of marine Synechococcus

Illumina sequencing of petB (cytochrome b6 subunit of the cytochrome b6f complex) marker gene from Synechococcus populations sorted by flow cytometry as well as collected by filtered seawater.Water samples were collected during TAN1810 oceanography campaign (21st October - 21st November 2018) at Southwest Pacific Ocean, near the Chatham Rise (NZ).Reads were obtained by:1) standard PCR with petB-F (TACGACTGGTTCCAGGAACG) and petB-R (GAAGTGCATGAGCATGAA) primers for filtered samples2) nested PCR with petB-F and petB-634R (GCTTVCGRATCATCARGA) during the first round and petB-50F (CAGGACATYGCTGAY) and petB-R during the second round of amplification, for flow cytometry sorted populations.
