Single-cell RNA-sequencing of spinal cord ependymal cells from FOXJ1-EGFP mice

The spinal cord neural stem cell potential is contained within the ependymal cells lining the central canal. Ependymal cells are, however, heterogeneous and we know little about what this reflects. To gain new insights into ependymal cell heterogeneity, we microdissected the ependymal cell layer from the thoracic spinal cord of 4 FOXJ1-EGFP transgenic mice (2.5-to-3-month old). After after dissociating the tissue into a cell suspension, we sorted single GFP-positive ependymal cells into lysis plates. cDNA synthesis was performed using Smart-seq2 technology.

脊髓神经干细胞潜能存在于衬覆中央管(central canal)的室管膜细胞(ependymal cells)内。然而,室管膜细胞具有异质性,我们对该异质性的内涵却所知甚少。为深入解析室管膜细胞异质性,我们从4只2.5至3月龄的FOXJ1-EGFP转基因小鼠的胸段脊髓中显微分离室管膜细胞层。将组织解离为细胞悬液后,我们将单个绿色荧光蛋白(Green Fluorescent Protein, GFP)阳性的室管膜细胞分选至裂解板中。随后采用Smart-seq2技术完成cDNA合成。
