The goal was to study the expression of the VL2-315 idiotope derived from MOPC315 in a novel mouse model. In the model, exon 2 of the germline VL2 gene was exchanged with a VL2-315 sequence with nucleotide changes that code for the idiotope in AA positions 94-96. Specifically, germline YST is converted to FRN. The VL2-315 modified mouse was designed to allow for physiological expression levels of VL2-315 and normal recombination of the targeted VL2-315 to JL and CL. To study the expression levels of VL2315 versus germline VL2, as well as to examine the junctional amino acid at position 98 of the V-J junction, we performed cDNA amplicon sequencing on splenic CD19+ B cells based on mVl1/2 and mCL-outer primers as published in Busse et al. EJI, 2014.
