This work aimed to evaluate the implications of pre-coating rat decellularized liver scaffolds with HepG2-derived conditioned medium (CM) on liver extracellular matrix quality. Our findings suggest that pre-coating improves recellularization, promoting HepG2, endothelial and mesenchymal cells adhension, spreading and proliferation. This proteomics shows LAMB1 and FN1 proteins retention in decellularized livers after HepG2-CM coating, as well as extracellular matrix organization and cell adhension proteins clusterization in coated livers interactome.

本研究旨在评估采用HepG2细胞来源的条件培养基(conditioned medium,CM)预包被大鼠脱细胞肝脏支架,对肝脏细胞外基质质量的影响。本研究结果显示,预包被可提升支架的再细胞化效率,促进HepG2细胞、内皮细胞与间充质细胞的黏附、铺展及增殖。蛋白质组学分析结果表明,经HepG2-CM包被的脱细胞肝脏中,LAMB1与FN1蛋白得以留存;同时,包被后肝脏的相互作用组中,存在细胞外基质组织相关蛋白与细胞黏附蛋白的聚集。
