资源简介:
Diabetes mellitus is one of the major independent risk factors for the development of ASCVD. Several studies have reported that diabetes increases inflammatary signaling in different cells, and the cytokines may influence plaque formation, but the signal pathway in circulating cells that links glucose metabolism and lipid metabolism remains unclear. We isolated peritoneal macrophages of diabetic and non-diabetic APOE-/- mice to investigate the diabetes influence on macrophages. APOE-/- mice were rendered diabetes from 8 weeks of age by intraperitoneal injections of streptozotocin (STZ, 40 mg/kg) or citrate buffer (nondiabetic) on 5 consecutive days, then the mice were fed with western diet for 8 weeks. In the 8th week, mice were i.p. with 1.5mL of 3% thioglycollate, and 3 days later, the peritoneal exudate was isolated for RNA isolation and sequencing.
