This dataset provides data from more than 8000 CA1 excitatory cells in mouse CA1, using a transgenic mouse line Thy1-GCaMP6s with two-photon calcium imaging in freely behaving mice with miniaturized microscopes. A total of 7 mice are included. Five of the mice underwent an experiment in which they freely foraged open field arenas of different colours (arena A and B), in a three-step paradigm of A-B-A meant to induce global remapping of CA1 place cells. Three of the mice underwent an experiment where they foraged arena A, being exposed to an object within the arena in a four-step paradigm of Empty-Object-Object moved-Empty, meant to show if cells in CA1 respond in relation to the introduced object. Only one mouse underwent both experiments. The mice were foraging for cookies on an 80x80 cm square enclosure, where box A and B had different colours and distal cues, and were located in different parts of the same room. The object introduced was a high-contrast duplo tower approximately 7x7x27 cm of size, unclimbable to the mice. Tracking was performed with an overstage camera and poses extracted using DeepLabCut. Calcium signals, motion correction and ROI detection was performed using suite2p. The data provides both high anatomical precision and high cell yields in freely behaving mice that might be of general interest.
本数据集以Thy1-GCaMP6s转基因小鼠品系为实验对象,通过微型化显微镜对自由活动小鼠进行双光子钙成像(two-photon calcium imaging),共采集其海马CA1区(CA1)超过8000个兴奋性神经元的相关数据。本实验共纳入7只小鼠:其中5只小鼠参与第一项实验,在A-B-A三阶段范式下自由觅食于颜色不同的开放场箱(箱A与箱B),该范式旨在诱导CA1位置细胞发生全局重映射;剩余3只小鼠参与第二项实验,在Empty-Object-Object moved-Empty四阶段范式下,先在箱A中自由觅食,随后依次向箱内放置物体、移动该物体、移除物体,该范式旨在探究CA1神经元是否会对引入的物体产生特异性响应。仅有1只小鼠同时参与了上述两项实验。所有实验小鼠均在80×80 cm的方形开放场箱中觅食饼干,箱A与箱B颜色及远端视觉线索各不相同,且放置于同一房间的不同区域。实验中引入的物体为尺寸约7×7×27 cm的高对比度乐高得宝(Duplo)积木塔,小鼠无法攀爬该物体。动物运动追踪通过箱顶摄像头完成,姿态提取采用DeepLabCut工具实现;钙信号处理、运动校正及感兴趣区(ROI,Region of Interest)检测均通过suite2p软件完成。本数据集在自由活动小鼠模型中同时实现了高解剖学定位精度与高神经元获取量,具备广泛的研究应用价值。
