We performed hepatocyte-specific knockout of Ccnd1 in 17-month-old C57BL/6J mice using AAV8-TBG-saCas9 with Ccnd1-targeting sgRNA versus Rosa26 control. After 3?weeks, livers were harvested for bulk RNA-seq to assess effects of elevated Ccnd1 with age. Overall design: Upon receipt, mice were rested for =2?weeks and maintained on normal chow. At 17?months, mice received retro-orbital AAV8-TBG-saCas9âU6-sgCcnd1 (3Ã10¹¹?vg/mouse) or Rosa26 control; livers (m-lobe) were harvested 3?weeks later for bulk RNA-seq.
