Intestinal epithelial O-GlcNAcylation not only responds to microbial cues but also in turn modulates the gut microbiota, which contributes to metabolic phenotypes observed in Vil-Ogt knockout mice. In the first experiment, we treated C57BL/6 mice with broad antibiotics to deplete gut bacteria, followed by the oral gavage of short chain fatty acids. In the second experiment, germ-free mice received fecal microbiota transplantation from wildtype or Vil-Ogt KO mice. Fecal DNA from these mice were extracted for Illumina MiSeq 300-bp paired-end sequencing of the V4 region of the bacterial 16S rRNA gene.
