QingGanJianPi oral solution ameliorates hyperlipidemia in C57BL/6 mice through metabolic mechanisms and intestinal flora

BACKGROUND: Hyperlipidemia, characterized by high blood lipid levels, is a high-risk factor and key indicator of many metabolic diseases, as well as a risk factor for many cardiovascular diseases. QingGanJianPi oral solution is a proprietary Chinese medicine clinically used to improve the complications of fatty liver. However, the ameliorative effect of QingGanJianPi Oral solution on hyperlipidemia has not yet been elucidated.METHODS: Pathological staining, WB, 16 S rRNA gene sequencing, and metabolomics were used in this study to determine whether with had an ameliorative effect on high-fat diet-induced hyperlipidemia in mice.RESULTS: The results showed that QingGanJianPi Oral solution and the combination of the two reduced the levels of total cholesterol (TC), triglyceride (TG), and low-density lipoprotein cholesterol (LDL-C) levels, and high-density lipoprotein cholesterol (HDL-C), and that QingGanJianPi Oral solution and Compound Mushroom Tablets attenuated high-fat diet-induced body weight gain and fat deposition, as well as modifying the microbial composition of the intestine. Non-targeted serum metabolomics identified relevant biomarkers to alleviate the overaccumulation of lipid droplets in the liver, while upregulating the ABCA1 signaling pathway , which is associated with and strongly correlated with hepatic lipid metabolism and phenotypic indicators.CONCLUSION: This suggests that QingGanJianPi Oral solution may act by modulating metabolic pathways. These results highlight its potential in the treatment of hyperlipidemia and provide direction for future research, including in-depth studies of the drug's bioactive components, dosage, and therapeutic strategies.
