The effects of adipose-derived stem cells (ADSCs) on the MCF-7 breast cancer cell line were investigated using a co-culture model followed by RNA sequencing. Co-culture of ADSCs and MCF-7 was conducted using a transwell plate, with MCF-7 cultured in standard conditions being used as controls. RNA extraction of MCF-7 cells (co-cultured and controlled) was performed using the TRIzol reagent kit (Thermo Fisher Scientific, USA). Messenger RNA was isolated using magnetic beads attached with oligo-T. Library preparation and paired-end sequencing were conducted by Novogene Co., LTD (Beijing, China) on the Illumina NovaSeq 6000 platform (San Diego, USA). Sequencing reads were 150 bases in length. Gene expression and functional enrichment analyses were then used to study the influences of ADSCs on MCF-7 cells at the transcriptomic level.
